About Glyphic:

At Glyphic Biotechnologies, we plan to create the protein revolution for which scientists and researchers have been waiting. We are developing a massively parallel, single-molecule proteome sequencing platform that will transform life science discovery and usher in a new era of insights into human biology and disease. To date, we have raised >$50M from venture partners and non-dilutive grant funding to achieve our vision of next generation proteome sequencing.

What we are looking for in you

We are seeking a technically driven, detail-oriented, and collaborative Senior/Staff Scientist to lead sample preparation and support assay development. In this pivotal role, you will report to the Director of Assay Development and work together to design, validate, and relentlessly optimize the core single-molecule protein sequencing platform, ProSE, ensuring it achieves industry-leading metrics for robustness, sensitivity, and commercial scalability. A critical component of this work involves developing and integrating specialized sample preparation protocols for diverse and challenging biological inputs. You will also collaborate closely with Chemistry to ensure these workflows seamlessly interface with the optimized ProSE platform, enabling a viable, high-throughput, end-to-end sequencing solution compatible with real-world samples.

The ideal candidate can independently troubleshoot complex technical challenges, rapidly iterate on experimental designs, and communicate effectively across multidisciplinary teams. Experience with high-throughput workflows, protocol adaptation for novel sample types, and driving projects from concept to implementation will separate the good from the great.

This role will have the unique opportunity to directly shape the future of single-molecule protein sequencing, enabling transformative discoveries in life science by making robust, scalable, and real-world-ready sequencing solutions a reality.

This is a full-time, exempt, in-person position with work conducted at our Berkeley, CA location.

What you’ll do

Assay Development:

  • Design, validate, and optimize ProSE (proprietary single molecule protein sequencing platform) experiments for robustness, sensitivity, and scalability, including improving reaction efficiency and kinetics in mixed phase assays.
  • Lead the integration of chemistry developments into assay applications, identifying and addressing potential incompatibilities.
  • Optimize and automate c-terminal and side chain blocking chemistries provided by other teams for use in the assay.
  • Propose and test high-sensitivity molecular tools to build predictive understanding of factors affecting assay robustness and sensitivity.

Sample Preparation:

  • Evaluate and process incoming KOL (Key Opinion Leader) samples, ensuring sample preparation methods accommodate specific constraints and requirements.
  • Assess diverse sample types, determine appropriate analytical approaches, and develop protocols for extraction and purification (cells, tissue), collaborating with chemists to scale and integrate into high-throughput workflows.
  • Own the introduction and validation of real biological sample inputs to enable sequencing of biologically relevant material, including complex and challenging samples.
  • Develop and implement strategies for sample enrichment, depletion, and dynamic range management to optimize assay performance.
  • Collaborate with early access KOLs to ensure seamless integration of sample workflows and address unique sample challenges.

What you need

Required:

  • Degree in biochemistry, chemical biology, or equivalent field. Advanced degree preferred.
  • 6+ years (PhD), 8+ years (MS), 10+ years (B.S.) of industry or equivalent experience in assay development.
  • Expertise in developing and optimizing single molecule or ultrasensitive molecular assays.
  • Deep understanding of high sensitivity highly multiplexed protein quantification (nELISA, NuLISA, immunoassays, PLE, PEA).
  • Expertise in developing sample prep or enrichment methods for low abundance targets or input material.
  • Experience working with real biological samples (non-contrived) and developing assays that account for matrix variability and real-world conditions.
  • Excellent problem-solving skills and attention to detail.
  • Excellent written and verbal communication skills, with the ability to work seamlessly with colleagues from diverse scientific and technical backgrounds.
  • Ability to thrive in a fast-paced environment with evolving priorities.

Nice to have:

  • Experience with nanopore platforms for sequencing or label free detection
  • Familiarity with nanoLC-MS based proteomic workflows and data analysis
  • Experience with single cell or spatial proteomics and transcriptomics

We’re looking for teammates with:

  • Excellent interpersonal skills – capable of building strong relationships and communicating effectively with stakeholders at all levels.
  • High emotional and analytical intelligence – able to navigate complex team dynamics, partnerships, and challenges with creativity and logic.
  • Resourceful adaptability – operates with urgency, remains flexible in evolving environments, and thrives in ambiguity.
  • Collaborative spirit – enjoys working across disciplines and explaining complex concepts to diverse audiences.

What you can expect from this role

Work environment:

  • Collaborative culture where your ideas and expertise are valued
  • Direct impact on product development and company direction

Professional growth:

  • Learn from a diverse team of world-class scientists and engineers
  • Contribute to first-of-their kind technologies, high-impact publications, and patents
  • Opportunity to develop and manage relationships with Key Opinion Leaders
